If it's been raining you will have residual water left in the tray. If they were blocked the water would be coming through the roof lining. A low pressure airline should clear them but if you use high pressure you could blow the drains off the pipes. Better to clear the actual blockage if you can find it. It sometimes clears with boiling water down the holes, but usually you need to physically push something down there. The drain holes will emerge in the wheel arches, so make sure you're using a long enough piece of wire. I have a Renault scenic and when i bought it i bought one with a solid roof because they are bad for filling up with rain water. Get a long length of stiff-ish wire and poke it down each hole. If it gets hard to push then you know there is a blockage.
